Abstract :研究成果の概要(和文):ニホンナシのみつ症は果肉が水浸状を呈する深刻な生理障害であり、その原因解明が急務となっている。そこで本研究では、‘みつ症が激しく発症する豊水’の四倍体を用いて、みつ症発症の原因となる遺伝子の探索を行った。マイクロアレイ解析の結果、四倍体では二倍体と比較し、みつ症発症前に細胞壁構成成分の代謝に関わる遺伝子や果実の成熟や老化に関わる遺伝子の強い発現を確認した。さらに詳細な発現解析を行ったところ、糖代謝に関わる2種類の遺伝子の発現量が、みつ症の発症前に四倍体果実において上昇することを確認した。これらの結果より、細胞壁や細胞質に存在する糖代謝に関連する遺伝子がみつ症の発症に強く関連すると考察した。研究成果の概要(英文): Water core of fruit is one of physiological disorder in Japanese pear. The disorder is very serious because a Japanese leading variety Housui is susceptible to it. Although the water core is thought a heritable character, their genetic mechanism is unknown. Recently, more sensitivity for water core had identified in Housui tetraploid than their diploid. In this research, we explored the candidate genes related to exhibit water core fruit in Japanese pear 'Housui' using microarray analysis between diploid and tetraploid. As a result of microarray analysis, some gene involving in fruit ripening, senescence and cell wall metabolism were strongly expressed in tetraploid fruit just before the exhibiting water core symptom. After analyzing more detailed DNA expression, we confirmed that two types of gene were highly expressed in tetraploid fruit before the symptom. We concluded that the candidate gene for sugar metabolism relate to the water core in Japanese pear.
