Title :昆虫におけるオーキシン生合成酵素の分子進化とゴール形成能獲得の因果性の解明

Abstract :研究成果の概要(和文):研究代表者らはこれまでにゴール形成昆虫が植物ホルモンの1種であるオーキシンを自ら合成し,高濃度に体内に保有していることを明らかにしていた。また,昆虫における独自の生合成経路を提唱し,それに対する阻害剤の取得も行った。本研究では,阻害剤を用いたオーキシン生合成のゴール形成への関与の立証,生合成経路の再検討,生合成酵素の同定を行った。その結果,昆虫体液に阻害剤の作用をキャンセルする物質が含まれており,個体レベルでオーキシン生産を抑えることが困難なこと,生成速度と代謝速度の差から生合成中間体が蓄積しない理由,主要な変換を担う生合成酵素などを明らかにした。 研究成果の概要(英文):We previously showed that galling insects biosynthesize auxin, a class of phytohormones, and accumulate it at high concentrations. We have also proposed the highly probable auxin biosynthetic pathway in insects, and have succeeded in obtaining a biosynthetic inhibitor. In this study, we tried to demonstrate the importance of auxin for gall induction by using the inhibitor, but it was clarified that some substances to cancel the function of the inhibitor exist in hemolymph, resulting in the failure in inducing auxin defect in insects in vivo. On the other hand, we have successfully interpreted the reason why biosynthetic intermediates cannot be detected by showing that their metabolizing rates were much faster than their producing rates. We have also successfully identified an enzyme involved in the conversion of indole-3-acetaldehyde to indole-3-acetic acid, the active auxin, from Bombyx mori silk-gland extract.
